It has been reported that approximately more than half of tumor patients require radiation therapy during the course of treatment. Clinically, the conventional mode of fractionated radiotherapy is 1.8~2.0 Gy/session. However, in recent years, with the advancement of radiotherapy technology and equipment, single higher dose fractionated radiotherapy (8~30 Gy) has attracted more and more attention from the radiotherapy community. According to the classical "4Rs principle" of radiobiology, macrofractional radiotherapy lacks redistribution within the cell cycle and reoxygenation of tumor cells. Therefore, macrofractional radiotherapy does not seem to have biological advantages from the perspective of traditional radiobiology. However, more and more clinical research results show that macrofractional radiotherapy has its own advantages in the treatment of solid tumors such as intracranial tumors and non-small cell lung cancer.
Connexins (Cxs) molecules are important components of intercellular gap junction, among which Connexin43 (Cx43) is the most abundantly expressed Cxs molecule. Studies have shown that tyrosine protein kinase Src can regulate the activity of Cx43 through various mechanisms, but the mechanism of how Src regulates Cx43 activity under radiation conditions, and the effects on gap junction-mediated irradiation side effects are not clear.
According to the latest research report, the incidence rate of malignant tumors in China is 235.23/100,000, and the mortality rate of malignant tumors is 148.81/10,000. The mortality rate of malignant tumors is increasing year by year, and it has become one of the leading causes of death for Chinese residents, accounting for about a quarter of the global cancer deaths. Radiotherapy has become one of the main treatment modes for malignant tumors. About 70% of patients with malignant tumors need radiotherapy (including comprehensive treatment and alone treatment). However, the resistance of cancer cells to radiation can lead to the failure of radiotherapy. Therefore, studying the molecular mechanism of intrinsic radiosensitivity of malignant tumors and seeking new molecular targets to predict individual tumor radiosensitivity are important ways to achieve radiosensitization and improve the effect of radiotherapy in tumor patients.
