Liver cancer is a common malignant tumor, 2018 world-renowned journalA Cancer Journal for CliniciansThe latest global cancer statistics published by (CA) show that liver cancer has the third highest mortality rate among all tumors. In the world, divided by regions, China belongs to the high incidence of liver cancer. For the treatment of liver cancer, surgical resection is the first choice for early detection. However, for most patients with advanced liver cancer who cannot be detected in time, surgical resection cannot be done due to excessive tumor size and tumor thrombus. For these patients, radiotherapy becomes the ideal treatment. Clinical studies have shown that radiotherapy can significantly improve the survival of patients with advanced liver cancer, but radiation-induced liver disease induced by radiotherapy restricts clinical radiotherapists from giving large doses of radiation irradiation. Therefore, at the current conventional dose tolerated by the liver, radiotherapy combined with radiosensitizing drugs is one of the ways to increase the effect of radiotherapy.
Vascular endothelial cells are very sensitive to ionizing radiation, and they are one of the important target cells for damage to various systems caused by radiation. Radiation damage of vascular endothelial cells is closely related to hematopoietic dysfunction, immunocompromise, gastrointestinal toxicity and other radiation damage manifestations. The lesions caused by vascular endothelial cell injury were observed in patients exposed to nuclear and radiation accidents, patients with clinical tumor radiotherapy and people in high background areas. Thrommodulin (Thbd) is a transmembrane glycoprotein located on the surface of vascular endothelial cells. It is not only a marker of radiation-induced vascular endothelial cell damage, but also an important drug target to alleviate radiation toxicity. Activation of Thbd signaling pathway helps to protect hematopoietic stem/progenitor cells, alleviate the radiation damage of vascular endothelial cells, and alleviate the inflammatory response caused by radiation.
