Evaluating the genotoxicity of exogenous compounds by reflecting chromosome abnormalities in the incidence of micronucleus (MN) has been widely used in the safety evaluation of compounds, ecological environment and biological monitoring^[1]。 At the same time, human peripheral blood lymphocyte micronucleus test is widely used in the investigation of environmental impact on human health, the monitoring of genetic damage in occupational exposure population, and the investigation of genetic damage in patients receiving chemotherapy. Compared with in vivo erythrocyte micronucleus flow detection and in vitro cell line micronucleus flow detection, there are few reports on human peripheral blood lymphocyte micronucleus flow detection, and the research mainly focuses on using flow detection of human peripheral blood lymphocyte micronucleus to evaluate the genotoxicity of exogenous compounds^[2]。 Flow cytometry used two fluorescent dyes to label cells, ethidium monazide (EMA) and SYTOX^©Green fluorescence (SYTOX^©Green). Differentiate the main nucleus from the micronucleus according to the intensity of green fluorescence^[3,4,5]。 The dose-effect curve and subject operating characteristic curve (ROC curve) of human peripheral blood lymphocyte micronucleus were established by using routine micro-blood culture micronucleus detection (Method A) as the standard, and peripheral blood mononuclear cell (PBMC) was isolated by culture flow micronucleus detection (Method B). To investigate the correlation between the detection of lymphocyte micronucleus by flow cytometry and the detection of micronucleus by routine culture and the possibility of applying flow cytometry to the health examination of peripheral blood lymphocyte micronucleus in radiation workers.

Brain metastasis from breast cancer (BCBM) is common in clinic, and its mechanism, characteristics, treatment and prognostic factors are complex^[1,2,3,4]。 Traditional whole brain irradiation (WBRT) has obvious curative effect in controlling symptoms^[5]。 However, in recent years, its benefit in patients with oligometastasis (number of brain metastases ≤3) or with a good graded prognostic assessment score (GPA) has been questioned. The reasons include the positive survival efficacy of lesions 1 to 3 alone or combined with surgery with stereotactic irradiation (SRS), the negative effects of WBRT on neurocognitive and emotional function and quality of life (QoL), and the insufficient evidence of WBRT improving survival of multiple brain metastases^{[6,7,8,9,10,11]}。 In recent years, effective systemic therapies (chemotherapy, endocrine, and targeted therapies) have prolonged the survival of stage IV breast cancer, including BCBM^[12,13,14]。 Considering that WBRT can reduce or slow the recurrence of brain tumors or the formation of new foci, combined systemic treatment of WBRT has the potential to independently improve BCBM overall survival (OS) and survival without intracranial disease progression (PFS). The aim of this study was to evaluate the independent relationship of WBRT and its dose grouping with OS and PFS through a retrospective multivariate Cox survival analysis.

cutaneous squamous cell carcinoma (cSCC, referred to as cutaneous squamous cell carcinoma for short) is the second most incidence cancer in the world. It is highly malignant and has strong metastatic ability. The 5-year survival time is only 22% ~56%, and there are more than 1 million new cases every year. Excessive ultraviolet radiation exposure is the primary environmental factor that induces skin squamous cell carcinoma. It is urgent to understand the molecular mechanism of ultraviolet-induced skin squamous cell carcinoma in depth and develop effective new therapies for skin squamous cell carcinoma. Hypoxia-inducible factor HIF-1 α is sensitive to ultraviolet radiation, and its expression level is closely correlated with the proliferation and glycolysis metabolism of skin squamous cell carcinoma cells, and is positively correlated with the grade of skin squamous cell carcinoma. However, previous studies have not revealed its regulatory mechanism.
